Over the last decade a new scientific area-ionomics began its development, the study ot the ionome. The ionome is the mineral nutrient and trace element composition of the organism, representing the inorganic component of the cellular and organismal system and the changes that occur in this composition in response to physiological and genetic modifications. The aim of the thesis is to propose a method for determination of large number of elements for characterization of the plant ionome and to assess changes in Taraxacum officinale (dandelion) ionome under different anthropogenic influence. Optimization of the analytical methodology and adequate choice of various measurement conditions (cold and hot plasma, reaction cell) were done for developing a highly accurate and precise method for determination of macro, micro and potentially toxic elements. As a result of the experimental work a method for the determination of 60 elements in plant and soil samples using quadropole ICP-MS is proposed and validated. The plant ionome of Taraxacum officinale collected from background and contaminated area was characterized. Both transfer and accumulation factors in various sampling sites were determined. The distribution of the elements in different plant parts of Taraxacum officinale was established. Possitive and negative correletions were observed among elements depending on the anthropogrnic influence. For the first time a method for determination of all 60 elements in dandelion for characterization of plant ionome is proposed which helps to reveal changes in the elemental concentrations of rarely determined elements in dependence on environment concentration. |
